The 2281S is only able to create a battery model using a charging cycle and cannot generate models for non-rechargeable batteries. Andrea's suggestion provides much more flexibility and simplicity using a 2450 0r 2460 series source meter.

I think the most efficient solution is to develop a UTM and code the SMU to change levels. Timing control will be somewhat inaccurate but you should be able to achieve reasonable results. You might want to try setting the Reading Delimiter to String Terminator. I believe the 4200 is expecting a comma terminator. This setting is likely a legacy setting.
